CYP1A2 and derivatives are responsible for metabolism of xenobiotics. Approximately 70% of xenobiotics, drugs and toxins, are biotransformed and removed by this group of enzymes. Altering the ability of an enzyme from this group to metabolize xenbiotics may increase the concentration and time these xenobiotics persist in the body potentially increasing the incidence of toxicity.
8-methylxanthine furafylline is a mechanism-based inhibitor of CYP P450 1A2 (CYP1A2). Oxidation of the C-8 methyl group by CYP1A2 results in a 1:1 adduct formation with an unknown amino acid in the active site of CYP1A2. Inhibition is irreversible. -{Racha 1998; Kunze 1993}-
