Mechanism of DNA Polymerase alpha inhibition by aphidicolin

Context

Aphidicolin,a tetracyclic diterpenoid, is a specific inhibitor of DNA polymerase alpha -{Glover 1984}-. Exposure to aphidicolin results in chromosomal aberrations -{Glover 1984}-.

Mechanism

Aphidicolin is a competitive inhibitor for pol alpha resulting in increased chromosomal aberrations. Aphidicolin forms a complex with DNA polymerase alpha resulting in inhibition of polymerase function causing chromosome gap and break formation -{Glover 1984}-.
