Is it even possible eg grna plot requires single guide? Do they expect 4 guides per KO? etc.

solution: make pseudo gRNA assignments e.g., grnaX-2_grnaY-4 becomes grnaXgrnaY-24 i.e., unique combination of two (or more) gRNAs for Mixscape.
